  • Dynamic expression lines. Wrinkles visible only during muscle contraction (frown lines, crow's feet while smiling). Respond to lower Snap-8 concentrations (0.5–2%) because the target is reducing the intensity of the muscle contraction itself. The peptide doesn't need to eliminate movement, just attenuate it enough that repetitive folding doesn't deepen the crease over time.
  • Static expression lines. Wrinkles visible at rest due to cumulative collagen damage. Require higher concentrations (3–5%) sustained over longer periods (12–16 weeks minimum). The mechanism here is indirect: by reducing the mechanical stress from repeated contraction, Snap-8 creates a recovery window where endogenous collagen remodelling can partially fill the existing crease. This is not the same as stimulating new collagen synthesis. Snap-8 lacks that mechanism. The improvement comes from removing the ongoing damage signal.
  • A third category. Mixed lines (visible at rest but significantly deeper during contraction). Benefits most from layered protocols: 2–3% Snap-8 for contraction attenuation combined with retinoids or ascorbic acid for collagen synthesis stimulation. The peptide reduces deepening; the actives address repair. Neither works optimally alone for this presentation.
  • Application timing matters because acetylcholine release follows circadian patterns. Peak facial muscle activity occurs during waking hours, meaning twice-daily application (morning and evening) maintains more consistent SNARE inhibition than once-daily dosing. Night-only application misses the highest-activity window entirely.
