Peptide mapping is a critical quality test to confirm the primary structure of proteins used in biotherapeutics such as monoclonal antibodies and antibody-drug conjugates. It relies on protein digestion which includes the enzymatic treatment of a protein to produce peptide fragments to further characterize specific regions and modifications. The results of protein digests are essential for protein structural characterization, protein identification, and protein modification monitoring including post-translation modifications, product related impurities, and multi-attribute monitoring (MAM) methods. However, sample preparation methods for peptide mapping are often complex, laborious, irreproducible, and require long digestion times—as much as 24 hours in some cases.
