Protocol of OVA Peptide 323-339
Adoptive Transfer With DCs and In Vitro Restimulation of T Cells[1]
This protocol only provides a guideline, and should be modified according to your specific needs.
1. Mature dendritic cells (DCs) were incubated with 10μg/ml OVA Peptide 323-339 at 37°C for 2h.
2. After being washed twice, the cells were resuspended in serum-free RPMI 1640 medium.
3. The treated mature dendritic cells (mDCs) (1×10^6) were intravenously injected into C57BL/6J mice on days 1, 3, and 5.
4. On day 8, the mice were euthanized, and their splenocytes were harvested and restimulated with 10μg/ml OVA Peptide 323-339 at 37°C for 3 days.
5. The splenocytes were washed with PBS, stained with FITC-conjugated anti-mouse CD4 antibody, and further analyzed by flow cytometry. The fluorescence intensity indicated the proliferation of CD4 T cells.
