As the coronavirus outbreak shows, viruses are a constant threat to humanity. Vaccines are regularly developed and deployed against specific viruses, but that process takes a lot of time, doesn't help everyone who needs protection, and still leaves people exposed to new outbreaks and new viruses.

Now, researchers at Massachusetts General Hospital (MGH) have uncovered a novel potential antiviral drug target that could lead to treatments protecting against a host of infectious diseases - creating a pan, or universal, treatment. Their work suggests that the protein Argonaute 4 (AGO4) is an "Achilles heel" for viruses.

AGO4 is one of a family of AGO proteins. Until now, there has been little evidence of why they are important. The researchers, led by Kate L. Jeffrey, PhD, and her collaborators found that AGO4 plays a key role protecting cells against viral infections.

Specifically, this protein is uniquely antiviral in mammalian immune cells. The group studied the anti-viral effects of several Argonaute proteins, and found that only cells that were deficient in AGO4 were "hyper-susceptible" to viral infection. In other words, low levels of AGO4 make mammalian cells more likely to become infected.

This study was published today by Cell Reports.

The MGH researchers suggest that boosting levels of AGO4 could shore up the immune system to protect against multiple viruses. "The goal is to understand how our immune system works so we can create treatments that work against a range of viruses, rather than just vaccines against a particular one," says Jeffrey.

Mammals have four Argonaute proteins (1-4), which act by silencing genes and which are remarkably conserved throughout multiple living things, including plants. These are RNAi and microRNA effector proteins and RNAi is the major antiviral defense strategy in plants and invertebrates. Studies of influenza infected mice have shown that AGO4-deficient animals have significantly higher levels of the virus.

However, if there is no deficiency, there is no clear evidence that supplements will make a difference. No research studies concluded that supplements treat or prevent age-related, natural hair damage or loss or lead to healthier skin. Two studies in the early 1990s did suggest that a biotin supplement may cause the hair to become stronger and strengthen weak nails. However, the studies were small and not reproduced.

RAS mutations are one of the most common cancer-causing genetic mutations, and the drug is now being studied in several cancer types. I think it's going to work especially well in tumors that are primarily RAS driven, including colon cancer and lung cancer. It might also work in other cancer types in combination with drugs targeting other genetic mutations, but further research is needed.

You must only eat a high-carb diet on the days you have a heavy workout, which uses up the calories.

05In safety assessment, the consequences of being wrong are significant. How do you think about uncertainty and model limitations for safety and discovery?

Uncertainty and model limitations are critical in safety assessment. There is clearly greater potential for animal or human harm from an incorrect safety assessment than from making the wrong molecule in early discovery, although uncertainty matters in both contexts. It is important to distinguish different types of uncertainty. Aleatoric uncertainty reflects irreducible randomness in a task, while epistemic uncertainty is model-related, such as hallucinations or lack of knowledge. For LLM systems, we use multiple methods to measure consistency and we have built fact-checking approaches in which a separate LLM independently verifies the primary engine's outputs. For QSAR models, including our SAR work around seizure liability, we can use statistical metrics such as confidence intervals to quantify and communicate uncertainty. Understanding that uncertainty exists and communicating it in both safety and discovery is critical. Uncertainty metrics tell us where potential blind spots may exist in our modelling allowing us to take action to improve over time. This applies to both safety and discovery. The aim is not to pretend AI removes uncertainty, but to make uncertainty visible so scientists can make better-informed decisions.
